DO IT HAVE A KEY LOCK OR BUTTON TO OPEN IT ?

It has a key to open it.If you need more keys call the manufacturer, they will send it to you.

Is there a way to set it up to open without requiring the key?

It doesn't seem so. We left the key in to make it easier to open.

do you just use the normal multifold 9.5 x 9.10 towels?

we use the Marathron Premium Multifolds which are 9.25L x 9.5w so your 'normal multifold' should work.

Can you see how many towels are in the dispenser?

2 packs of 250 towels/pack. (depending on thickness of paper) per pack of 250 is approx. 5 inches. I hope this helps you
