Can I change one antenna to a directional antenna and use in repeater mode?

You could change the antenna, the antenna is uFL to RP-SMA-Male.

If I connect an external antenna do I have to have something connected to both antenna ports and if so do they have to match or can I leave one empty?

Coverage will likely be better with two similar antennas connected. One antenna connected would not, most likely, cause any circuit failures. GL.iNet has indicated to me that dissimilar antennas is acceptable. In my case I tried a WiFi yagi on one antenna port (supplied antenna on the other) but it did not improve signal strength to my internet source in my particular situation. I would suggest you give it a try and see if it works for you.
