With the normal door handle holes. Will this lock fit into it? Or must you need to make new holes?

Nope, you’ll need to either expand out the already present hole via either just winging it and checking to make sure the bolt mechanism has leeway to operate or use the provided templates, or drill a entirely new hole into the door

can you lock the door while you are in from the inside

Yes. On the inside you turn the handle to lock it. You have to, it doesn’t lock automatically.

Will this be able to be fitted onto a 40mm wood door? Intending to use it above a standard internal door handle

i don't have it any more but i think it can i had it on a think fire door just fine and i think that was about 40mm.
