Yes, in fact, as far as it is known, up to 4 players can play this game on the same tv so long as they all have a controller.

Q: What controllers are compatible with the game?

Along with using the 3DS copy of Smash Brothers to control the game, if you don't have a copy of the 3DS version, you can purchase the Smash Controller from the 3DS eshop for 99 cents.
