No, you can't customize the letters they come as a set, so if you need multiple, you need to order more sets.

Q: Can these be applied to nylon, such as a bomber jacket? Do you recommend securing with fabric glue ?

It can be pasted and fixed with E6000 glue, AB glue, glue stick, etc.

Q: Can you add additional glue for instance, clear Gorilla Glue that's water 💧 proof/safe for the water? Gratitude for your time!♡

The letters I received were very sticky, but I don’t know if they were waterproof sticky. If you’re going to use Gorilla glue to make them waterproof, I would put the glue directly on the item you’re putting the letters on then place the letter on top if that.
